Luise wis dochter o Prince George William o Hesse-Darmstadt (1722–1782) frae his marriage tae Coontess Maria Louise Albertine o Leiningen-Falkenburg-Dagsburg (1729–1818) who wis dochter o Count Christian Karl Reinhard of Leiningen-Dachsburg-Falkenburg-Heidesheim.
Frae 1770 the princess wis pally wi Marie Antoinette. Luise exchanged letters with the French queen tae 1792 when she wis cowpt.
Luise hersel married on 19 February 1777 in Darmstadt tae her cousin the hereditary prince Louis I of Hesse-Darmstadt (1753–1830). Her husband ruled Hesse-Darmstadt frae 1790 as Landgrave Louis X an frae 1806 as Ludwig I, Grand Duke of Hesse and the Rhine.
